The citrullinated EBV antigen assay quantitates citrullinated IgG antibodies in human samples such as serum. The assay supplements currently existing CCP assays and presents with high assay sensitivity and specificity. 20-30% of conventional CCP negative reactions are tested positive in this assay.
Human samples, standards and controls are added to the wells of microtiter wells precoated with citrullinated EBNA2 peptide and bind to the immobilized peptide. An enzymatic conjugated goat anti human IgG is added to each well, which binds to the antibody-peptide complex. A substrate is added that reacts with the enzyme-labelled target complex and a colorimetric measurable signal is produced. The intensity of the colorimetric reaction is directly proportional to the concentration of citrulline-dependent IgG antibodies in the analysed sample.
